PT-141 is the research name for bremelanotide, a peptide that acts on the brain to increase sexual desire. Brand-name Vyleesi, an injection used as needed, was FDA-approved in 2019 for premenopausal women with acquired, generalized hypoactive sexual desire disorder (HSDD). Telehealth providers also prescribe compounded PT-141 injections or nasal sprays, including for men, which is off-label.
How it works
Unlike ED medications such as sildenafil, which increase blood flow, bremelanotide works in the brain. It activates melanocortin receptors (mainly MC4R) in areas involved in sexual desire and arousal. In Vyleesi's phase 3 trials in premenopausal women, it produced small but statistically significant increases in desire and reductions in distress compared with placebo; it did not significantly increase the number of satisfying sexual events.
