How PT-141 (bremelanotide) Works

Unlike ED medications such as sildenafil, which increase blood flow, bremelanotide works in the brain. It activates melanocortin receptors (mainly MC4R) in areas involved in sexual desire and arousal. In Vyleesi's phase 3 trials in premenopausal women, it produced small but statistically significant increases in desire and reductions in distress compared with placebo; it did not significantly increase the number of satisfying sexual events. Studies in men, mostly earlier and smaller, suggested it can help erections, including in some men who don't respond to sildenafil, but it is not approved for men.

What to Expect

Taken as needed, not daily. Vyleesi is injected under the skin at least 45 minutes before sexual activity, and the label limits use to one dose per 24 hours and eight doses per month. Effects can last several hours. Women who see no improvement after 8 weeks of trying it should stop, according to the label.
