Sermorelin is a synthetic analogue of growth hormone-releasing hormone (GHRH) that stimulates the pituitary gland to release the body's own growth hormone. It was previously FDA-approved as Geref for diagnosing and treating growth hormone deficiency in children; the manufacturer discontinued it in 2008 for commercial reasons, not safety. Today it is available only as a compounded injection, prescribed off-label for adult body composition, sleep, and recovery.
How Sermorelin Works
Sermorelin consists of the first 29 amino acids of natural GHRH, the active portion of the hormone. It binds GHRH receptors in the pituitary and triggers pulsatile growth hormone release, which in turn raises IGF-1 from the liver. Because it works through the pituitary, the body's normal feedback (somatostatin) still limits GH output — a theoretical safety advantage over injecting growth hormone directly. Its half-life is very short, so it is usually injected nightly to coincide with natural GH pulses during sleep. Evidence for adult anti-aging use comes from small, older studies showing IGF-1 increases and modest body-composition changes; there are no large modern trials.
What to Expect
Typically injected subcutaneously once daily at bedtime, sometimes on a 5-days-on, 2-days-off schedule. Some patients report sleep changes within a few weeks.
